目的: 建立重组人胰岛素V8 酶肽图谱的毛细管电泳分析方法。方法: 利用毛细管区带电泳分离模式, 采用熔凝石英毛细管柱, 检测了重组人胰岛素经V8 蛋白酶酶解所产生的5 个肽片段;比较了重组与天然人胰岛素的V8 蛋白酶肽图谱; 鉴定了毛细管区带电泳和反相高效液相色谱的肽图谱中各峰的对应关系。结果: 重组人胰岛素与天然人胰岛素的分子结构具有同一性; 毛细管区带电泳和反相高效液相色谱2 种方法具有不同的分离机理。结论: 毛细管区带电泳可以成为反相高效液相色谱的有效补充用于重组人胰岛素的常规鉴定。
Objective: To establish a capillary electrophoresis method for the analysis of recombinant human insulin V8 peptide map. METHODS: Five peptide fragments produced by the enzymatic hydrolysis of recombinant human insulin by V8 protease were detected by capillary zone electrophoresis separation mode using fused silica capillary column. The V8 protease peptide map of recombinant and native human insulin was compared. Correspondence between peaks in peptide maps by capillary zone electrophoresis and reversed-phase high performance liquid chromatography. Results: The molecular structures of recombinant human insulin and native human insulin were identical. The two methods of capillary zone electrophoresis and reversed-phase high performance liquid chromatography had different separation mechanisms. Conclusion: Capillary zone electrophoresis can be an effective complement to RP-HPLC for the routine identification of recombinant human insulin.
