为建立肿瘤细胞学新的研究方法,本文应用培养的胰腺癌细胞通过细胞间隙连接通讯功能测定技术,将细胞以CFDA/AM染色后,在37℃及5%CO2内孵育10分钟,再经激光扫描共聚焦显微镜进行荧光测定。结果发现胰腺癌细胞的荧光恢复速率明显上升(0152±0117比98±0247,P<001),表明细胞间隙连接通讯功能迅速增强,细胞增殖受到抑制。本研究提示新的监测方法为检测基因治疗效果提供一项重要指标,也为肿瘤细胞生物学的研究提供了新的手段。
In order to establish a new research method of tumor cytology, the cultured pancreatic cancer cells were stained by CFDA / AM and then incubated in 37 ℃ and 5% CO2 for 10 minutes. Scan confocal microscopy for fluorescence measurements. The results showed that the fluorescence recovery rate of pancreatic cancer cells was significantly increased (0152 ± 0117 vs 98 ± 0247, P <001), indicating that the cell gap junctional communication function increased rapidly and cell proliferation was inhibited. This study suggests that the new monitoring method provides an important indicator for detecting the effect of gene therapy and also provides a new means for the study of tumor cell biology.
