目的探讨转移性乳腺癌组织中微小核糖核苷酸-200c(miR-200c)和过氧化物酶3(PRX3)表达水平联合预测保乳术后患者预后情况的价值。方法选取2010年1月~2012年1月在该院行保乳术治疗的转移性乳腺癌患者63例为研究对象,取其乳腺癌组织(乳腺癌组)及癌旁正常组织(正常组),采用免疫组化法检测两组miR-200c和PRX3表达水平。患者术后均行放化疗综合治疗,统计患者3年生存率,分析讨论转移性乳腺癌组织中miR-200c和PRX3表达水平与患者3年生存率的关系及两者联合预测患者预后情况的价值。结果乳腺癌组中miR-200c和PRX3蛋白阳性表达率均高于正常组,差异有统计学意义(P<0.05)。63例转移性乳腺癌患者3年生存率为46.03%,且存活患者miR-200c和PRX3蛋白阳性表达率均低于死亡患者(P<0.05)。Spearman秩相关分析结果显示,miR-200c和PRX3蛋白阳性表达率与患者3年生存率呈负相关(r=-0.775、-0.753,P<0.05)。ROC曲线分析结果显示,miR-200c和PRX3蛋白阳性表达率联合预测转移性乳腺癌患者保乳术后预后情况的价值良好,其中截断值为69.84%+63.49%时可同时取得较高的敏感度和特异度。结论转移性乳腺癌组织中miR-200c和PRX3蛋白阳性表达率较高且两者联合预测患者预后的价值良好,这可能与miR-200c和PRX3促进乳腺癌细胞侵袭转移相关,miR-200c和PRX3蛋白阳性表达率可用于转移性乳腺癌保乳术预后评估的参考指标。
Objective To investigate the relationship between the expression of miR-200c and PRX3 in metastatic breast cancer to predict the prognosis of patients after conservative surgery. Methods Sixty-three patients with metastatic breast cancer undergoing conservative surgery from January 2010 to January 2012 were enrolled in this study. Breast cancer (breast cancer) and adjacent normal tissues (normal) Immunohistochemistry was used to detect the expression of miR-200c and PRX3 in both groups. The patients were treated with radiotherapy and chemotherapy comprehensively and the 3-year survival rate was calculated. The relationship between the expression of miR-200c and PRX3 and the 3-year survival rate of patients with metastatic breast cancer was analyzed and discussed, and their prognostic value was evaluated . Results The positive rates of miR-200c and PRX3 in breast cancer group were significantly higher than those in normal group (P <0.05). The 63-year survival rate of 63 patients with metastatic breast cancer was 46.03%, and the positive rates of miR-200c and PRX3 protein in survival patients were lower than those in patients with death (P <0.05). Spearman rank correlation analysis showed that the positive rates of miR-200c and PRX3 were negatively correlated with 3-year survival (r = -0.775, -0.753, P <0.05). The results of ROC curve analysis showed that the combination of miR-200c and PRX3 protein positive predictive value in prognosis of breast-conserving patients with metastatic breast cancer was good, and the cut-off value of 69.84% + 63.49% And specificity. Conclusions The positive expression rate of miR-200c and PRX3 in metastatic breast cancer tissues is high, and the combination of them predicts the prognosis of patients with breast cancer. This may be related to the promotion of invasion and metastasis of breast cancer cells by miR-200c and PRX3.
