目的了解2010-2012年贵州省H3N2亚型流感病毒HA1基因变异特征。方法用MDCK细胞分离流感病毒,采用血凝抑制试验分型鉴定,选取23株H3N2亚型流感毒株进行RNA提取,逆转录扩增获得HA1基因产物,将其进行基因产物测序,测定的序列用相关的生物信息软件进行分析。结果贵州省2010年及2012年H3N2亚型流感病毒流行株与WHO疫苗推荐株A/Perth/16/2009(H3N2)亲缘关系较远,以至于H3亚型在我省成为优势株,引起广泛流行。结论 2010年及2012年H3N2亚型流感病毒流行株与WHO推荐的疫苗株之间的匹配性不完全一致。同时在2010-2012年间贵州省H3N2亚型流感病毒有持续的变异。
Objective To understand the variation of HA1 gene of H3N2 influenza virus in Guizhou province from 2010 to 2012. Methods The influenza virus was isolated from MDCK cells and identified by hemagglutination inhibition assay. 23 strains of H3N2 influenza virus were selected for RNA extraction and HA1 gene product was obtained by reverse transcription amplification. The products were sequenced. Related bioinformatics software for analysis. Results The prevalence of H3N2 subtype influenza viruses in Guizhou Province in 2010 and 2012 was far from that of WHO vaccine recommendation A / Perth / 16/2009 (H3N2), so that H3 subtype became the predominant strain in our province and caused widespread epidemic . Conclusion The match between H3N2 subtype influenza virus strains and WHO recommended vaccine strains in 2010 and 2012 is not consistent. In the meantime, H3N2 subtype influenza virus in Guizhou Province has a continuous variation in 2010-2012.
