目的 研究糖尿病(DN)大鼠模型经静脉注射PI3K抑制剂雷帕霉素后肝组织白介素-1(IL-1)与白介素-6(IL-6)表达的变化.方法 成年健康清洁级SD雄性大鼠60只,随机分为对照组(20)、糖尿病组(20)和雷帕霉素组(20).采用高脂饲料喂养和小剂量链脲佐菌素(STZ)腹腔内注射方法,建立2型糖尿病合并非酒精性脂肪肝动物模型.评价各组大鼠血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、三酰甘油(TG)、总胆固醇(CHO)和低密度脂蛋白胆固醇(LDL-C)的水平,采用免疫组织化学方法和Western blot方法检测三组大鼠肝组织IL-1与IL-6蛋白的表达.结果 糖尿病组大鼠血清ALT、AST、TG、CHO、LDL-C水平比对照组显著升高,而雷帕霉素组比糖尿病组显著降低(P<0.01).糖尿病组大鼠肝组织IL-1与IL-6蛋白的阳性表达量比对照组显著升高,而雷帕霉素组比糖尿病组显著降低(P<0.01).结论 雷帕霉素改善2型糖尿病大鼠肝功能可能与下调IL-1与IL-6的表达相关.“,”Objective To study the effect of rapamycin on the expressions of interleukin-1(IL-1) and interleukin-6(IL-6) in liver of the diabetic rat.Methods 60 healthy male rats were selected and randomly divided into the control group(20),the diabetic group(20)and rapamycin group(20).Rat model of type 2 diabetes mellitus with nonalcoholic fatty liver disease was established by intraperitoneally injecting small dose of chain urea with cephalosporins (STZ) and feeding high fat fodder.The serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG),cholesterol(CHO),low density lipoprotein-cholesterol(LDL-C) in serum were evaluated.The expressions of IL-1 and IL-6 protein in liver were observed by immunohistochemistry and Western blot methods.Results The serum levels of ALT,AST,TG,CHO,LDL-C were increased significantly in diabetic group than in control group,but decreased significantly in rapamycin group than in diabetic group(P<0.01).The expression levels of IL-1 and IL-6 proteins in lever were increased significantly in diabetic group than in control group,but decreased significantly in rapamycin group than in diabetic group(P<0.01).Conclusion The protective effect of Rapamycin on the liver of type 2 diabetic rats might be related to down-regulating the expressions of IL-1 and IL-6 in liver.
