目的:测定中药白胡椒中胡椒碱的含量。方法:采用高效毛细管电泳法,电泳条件为石英毛细管柱(50μm×90cm,实际长度81.5cm),运行缓冲液为0.02mol/LNa2HPO4:0.02mol/LNaH2PO4=95:5(16℃时pH=8.6),分离电压为28kV,压力进样30mbar×8s,温度为25℃,检测波长为343nm。结果:胡椒碱在0.05~0.30mg/mL范围内呈良好的线性关系(r=0.9995),平均回收率分别为99.82%,99.95%,100.20%;RSD分别为0.30%,0.69%,0.29%(n=3)。结论:高效毛细管电泳法可用于测定白胡椒中胡椒碱的含量。
Objective: To determine the content of piperine in Chinese white pepper. Methods: Capillary electrophoresis was used. The electrophoresis conditions were quartz capillary column (50μm×90cm, actual length 81.5cm). The running buffer was 0.02mol/LNa2HPO4: 0.02mol/LNaH2PO4=95:5 (pH=8.6 at 16°C). The separation voltage was 28 kV, the pressure was sampled at 30 mbar×8 s, the temperature was 25° C., and the detection wavelength was 343 nm. Results: There was a good linear relationship (r=0.9995) in the range of 0.05-0.30 mg/mL for piperine. The average recoveries were 99.82%, 99.95%, and 100.20%, respectively; the RSDs were 0.30%, 0.69%, and 0.29%, respectively ( n=3). Conclusion: High performance capillary electrophoresis can be used to determine the content of piperine in white pepper.
