目的探讨冬虫夏草口服液对焦虑小鼠行为干预效果及其作用机制。方法雄性昆明种小鼠40只随机平均分为生理盐水组、地西泮组、低剂量虫草组和高剂量虫草组。地西泮组前3天灌胃0.9%NaCl溶液,后4天灌胃地西泮(5mg.kg-1.d-1);低、高剂量虫草组分别连续灌胃7天不同剂量(0.75g.kg-1.d-1,7.5g.kg-1.d-1)的冬虫夏草口服液。生理盐水组连续灌胃7天等容积的0.9%NaCl溶液(1ml)。所有实验组小鼠7天后开始进行高架十字迷宫检测,完毕后即刻处死检测血清中的GABA含量。结果高架十字迷宫实验中各药物组与生理盐水组比较,地西泮组与高剂量虫草组在开臂活动时间、距离和穿梭时间上差异均具有统计学意义(P<0.01);高剂量虫草组与低剂量虫草比较差异有统计学意义(P<0.01)。与生理盐水组比较,地西泮组及高剂量虫草组的血清GABA水平差异有统计学意义(P<0.01);与低剂量虫草组比较,高剂量虫草组血清GABA水平差异有统计学意义(P<0.05);与地西泮组比较,高剂量虫草组血清GABA水平差异有显著统计学意义(P<0.05)。结论冬虫夏草口服液抗小鼠焦虑作用明显,其可能通过调节体内主要抑制性氨基酸GABA的水平,发挥类苯二氮卓类药抗焦虑的作用。
Objective To investigate the effects of Cordyceps oral solution on behavioral intervention in anxiety mice and its mechanism. Methods Forty male Kunming mice were randomly divided into normal saline group, diazepam group, low dose Cordyceps group and high dose Cordyceps group. 0.9% NaCl solution was given to the diazepam group 3 days prior to intragastric administration of diazepam (5 mg.kg-1.d-1) 4 days later. The rabbits in the low and high dose groups were treated with different dosages of 0.75 g.kg-1.d-1, 7.5g.kg-1.d-1) Cordyceps oral solution. The saline group was orally administered with an equal volume of 0.9% NaCl solution (1 ml) for 7 days. After 7 days, all the experimental mice started to detect the elevated plus-maze, and immediately after sacrifice, the serum levels of GABA were determined. Results Compared with saline group, there was significant difference in the time, distance and shuttle time between open-arm activity and diazepam group (P <0.01); the high-dose Cordyceps Group and low dose Cordyceps difference was statistically significant (P <0.01). Compared with saline group, serum GABA levels in diazepam group and high dose Cordyceps group were significantly different (P <0.01). Compared with low dose Cordyceps group, serum GABA level in high dose Cordyceps group was significantly different (P < P <0.05). Compared with diazepam group, the serum GABA level of high dose Cordyceps group had significant difference (P <0.05). Conclusion Cordyceps oral anti-mouse anxiolytic effect is obvious, it may by regulating the body’s main inhibitory amino acid GABA levels, benzodiazepines play an anti-anxiety effect.
