目的建立1种同时测定腐竹中碱性嫩黄O和碱性橙Ⅱ的高效液相色谱方法。方法采用乙腈-水(7+3)溶液对粉碎后的腐竹样品进行超声提取,在甲醇和乙酸铵水溶液梯度条件下进行洗脱,并在436 nm波长下进行测定。结果该方法测定碱性嫩黄O和碱性橙Ⅱ在0.1~25.0μg/mL浓度范围内呈良好线性关系,相关系数分别为0.999 2和0.998 9,3个水平(5.0、10.0、15.0 mg/kg)的加标回收率分别为89.5%~93.6%和90.5%~93.9%,相对标准偏差分别为0.50%~1.03%和1.12%~1.36%,检出限碱性橙Ⅱ为0.015 mg/kg、碱性嫩黄O为0.010 mg/kg。结论此方法具有简单、快速、准确等优点,适用于腐竹中碱性嫩黄O和碱性橙Ⅱ的同时测定。
Objective To establish a high performance liquid chromatographic method for the simultaneous determination of Alkaloids O and Alkali Orange Ⅱ in yuba. Methods The milled yam samples were extracted with acetonitrile-water (7 + 3) solution, and eluted with a gradient of methanol and ammonium acetate aqueous solution at 436 nm. Results The method was linear in the concentration range of 0.1 ~ 25.0μg / mL for alkaline yellow O and alkaline orange Ⅱ with correlation coefficients of 0.999 2 and 0.998 9, respectively (5.0, 10.0 and 15.0 mg / kg ) Were 89.5% -93.6% and 90.5% -93.9%, respectively. The relative standard deviations were 0.50% -1.03% and 1.12% -1.36% respectively. The detection limit of alkaline orange II was 0.015 mg / kg, Alkaline yellow O is 0.010 mg / kg. Conclusion This method is simple, rapid and accurate. It is suitable for the simultaneous determination of basic yellow O and alkaline orange Ⅱ in yuba.
