目的考察温性经筋通贴膏的体外释放和透皮特性。方法采用改良的Franz扩散池装置,离体SD大鼠皮肤作为透过介质,以20%乙醇为接收液,采用双波长HPLC法同时测定接收液中补骨脂素、异补骨脂素、盐酸小檗碱的含量。结果补骨脂素、异补骨脂素、盐酸小檗碱24 h累计释放百分率分别为:47.58%、29.16%、31.64%;平均释放速率分别为6.529 2,10.730 9,7.253 2μg·cm~(-1)·h~(-1);补骨脂素、异补骨脂素24 h累计透过百分率分别为:6.83%,3.81%;平均透皮速度分别为1.867 4μg·cm~(-1)·h~(-1),0.695 9μg·cm~(-1)·h~(-1);促渗剂阴性贴膏中补骨脂素的透皮速率明显降低,异补骨脂素则未检出。结论温性经筋通贴膏中补骨脂素、异补骨脂素、盐酸小檗碱体外释放和透皮行为均符合Higuchi方程;处方中的冰片和氮酮对补骨脂素和异补骨脂素均有促渗作用。
Objective To investigate the in vitro release and transdermal properties of Wenjingjingjin paste. Methods A modified Franz diffusion cell device was used. The skin of isolated SD rats was used as permeation medium and 20% ethanol as the receiving solution. The simultaneous determination of psoralen, isopsoralen, hydrochloric acid Berberine content. Results The cumulative release rates of psoralen, isopsoralen and berberine in 24 h were 47.58%, 29.16% and 31.64%, respectively. The average release rates were 6.529 2, 10.730 9 and 7.253 2 μg · cm ~ ( -1) · h ~ (-1). The accumulative permeation percentages of psoralen and isopsoralen in 24 h were 6.83% and 3.81%, respectively. The mean transdermal rates were 1.867 4 μg · cm -1 ) · H ~ (-1) and 0.695 9μg · cm ~ (-1) · h ~ (-1) respectively. The transdermal rate of psoralen in the penetration enhancer-negative plaster was significantly decreased, while that of isopsoralen not detected. Conclusions The in vitro release and transdermal behaviors of psoralen, isopsoralen and berberine in Wenjing Jingjin Tong paste are in accordance with the Higuchi equation. The prescriptions of borneol and azone are effective for psoralen and heteropodia Psoralen have a role in promoting permeability.
