目的:探讨舒郁颗粒对抑郁模型大鼠海马神经元细胞凋亡的保护作用.方法:将雄性SD大鼠随机分为正常组、模型组、中药组三组.采用慢性不可预见性应激刺激(CUMS)结合孤养方式复制抑郁大鼠模型,通过称重、敞箱实验与糖水偏爱度等检测大鼠行为学改变.应用免疫组化与RT-PCR检测大鼠海马区转录因子环磷酸腺苷反应元件结合蛋白(CREB)及基因的表达水平.结果:与模型组相比,中药组在给药后体质量增加快(P<0.05 ); 与模型组相比,中药组的水平运动与垂直运动得分和糖水偏爱度均明显升高 (P<0.05).与正常组相比,模型组大鼠海马区CREB蛋白和mR-NA表达明显下调 (P<0.01); 与模型组相比,中药组海马区CREB蛋白和mRNA表达明显上调 (P<0.05).结论:舒郁颗粒抗抑郁的作用机制可能与其促进海马区 CREB蛋白与mRNA的活性和表达,进而减少神经细胞凋亡有关.“,”OBJECTIVE: To investigate the protective effect of Shu Yu granule on cell apoptosis of hippocampal neurons in rat model with depression.METHODS: Male SD rats were randomly divided into three groups: normal group,model group and TCM group.To use the chronic unpredictable stress stimuli ( CUMS) combineing with solitary raising replicated the rat model of depression.The rat behavior were tested by weight,open-field experiment.To use immunohistochemistry and RT-PCR detected rat hippocam-pus transcription factor cyclic AMP response element binding protein CREB and the expression levels of genes.RESULTS: Compared with the model group,TCM group after administration body mass increased faster ( P<0.05); compared with the model group,hori-zontal movement and vertical movement and sucrose preference scores were significantly higher in TCM group (P<0.05).Compared with the normal group,CREB protein and mRNA expression in rat hippocampus in model group were significantly lower (P<0.05);compared with the model group,CREB protein and mRNA expression were significantly increased in the hippocampus medicine group (P<0.05).CONCLUSION: The mechanism of action of antidepressant Shu Yu Grain may be related to the promotion of the activity and expression of CREB protein and mRNA in the hippocampus so as to reduce neuronal apoptosis related.
