目的:探讨p53基因产物在乳腺不典型增生和乳腺癌中的变化及其意义。方法:用ABC免疫组化法检测正常乳腺、乳腺囊性增生病不典型增生和乳腺癌细胞p53基因产物表达。结果:正常乳腺上皮细胞和乳腺囊性增生病上皮增生Ⅰ级者均无p53蛋白表达。不典型增生Ⅱ、Ⅲ级和乳腺癌组织中p53蛋白染色阳性率分别为10%、30%和35%。结论:本研究结果提示该基因的异常改变可发生于正常细胞经不典型增生向癌细胞转化期中,是乳腺上皮细胞向恶性转化的标志之一。
Objective: To investigate the changes and significance of p53 gene product in breast atypical hyperplasia and breast cancer. METHODS: ABC immunohistochemistry was used to detect p53 gene product expression in normal breast, atypical hyperplasia of breast cystic hyperplasia, and breast cancer cells. Results: There was no p53 protein expression in normal breast epithelial cells and epithelial hyperplasia I of breast cystic hyperplasia. The positive rates of p53 protein staining in atypical hyperplasia II, III, and breast cancer tissues were 10%, 30%, and 35%, respectively. Conclusion: The results of this study suggest that abnormal changes in this gene may occur during the transition from normal to atypical hyperplasia to cancer cells and are one of the hallmarks of malignant transformation of mammary epithelial cells.
