在使用单克隆抗体鉴定肿瘤相关糖结合抗原的性质过程中,最近发现了一种抗结直肠腺癌细胞株的单克隆抗体(GA50),称之为肿瘤相关糖抗原CA50。CA50抗原以神经节糖苷和唾液糖蛋白这两种形式存在,而且已从各种组织的原发癌和转移癌中分离出来。非上皮肿瘤及正常成人组织中(除胰腺外)均未发现此种抗原。因此CA50可能是各种癌的一种相关抗原。 用一种简单的放射免疫测量法,探测释放入血清的癌相关抗原CA50。 非癌症患者和对照组CA50值;150名健康对照者的CA50单克隆抗体的抑制率为0~25%((?)±SD=0.07±0.06),仅一人超过该值。肉瘤,恶性黑色素瘤,溃疡性结肠炎、肺炎的抑制率除极少数例外,均未超过25%。28例溃疡性结肠炎病人中仅2例>25%。
In the process of identifying the properties of tumor-associated carbohydrate-binding antigens using monoclonal antibodies, a monoclonal antibody (GA50) against colorectal adenocarcinoma cell lines was recently discovered, which is referred to as tumor-associated carbohydrate antigen CA50. The CA50 antigen exists in both ganglioside and salivary glycoprotein and has been isolated from primary and metastatic carcinomas of various tissues. No such antigens were found in non-epithelial neoplasms and normal adult tissues (except for the pancreas). Therefore, CA50 may be a related antigen of various cancers. The serum-associated cancer-associated antigen CA50 was detected by a simple radioimmunoassay. CA50 values for non-cancerous patients and controls; CA50 monoclonal antibody inhibition for 150 healthy controls was 0-25% ((?)±SD=0.07±0.06), and only one person exceeded this value. The exception rate for sarcoma, malignant melanoma, ulcerative colitis, and pneumonia was less than 25%. Of the 28 patients with ulcerative colitis, only 2 were >25%.
