目的:建立芪苓益肝颗粒中黄芪甲苷的含量测定方法。方法:采用高效液相色谱-蒸发光散射检测(HPLC-ELSD)法定量分析芪苓益肝颗粒中黄芪甲苷的含量,色谱柱Aglient Eclipse XDB C18(4.6 mm×250 mm,5μm),以乙腈-水(35∶65)为流动相,流速1.0 mL·min-1,柱温25℃;蒸发光散射(ELSD)检测器,漂移管的温度110℃,载气流速1.6 L·min-1。进样量10μL。结果:黄芪甲苷在0.498~9.96μg呈良好的线性关系,r=0.9998;平均回收率为98.05%,RSD 0.89%,在常温下8 h内稳定性较好。结论:HPLC-ELSD法简便快速,准确,重复性好,结果可靠,可作为芪苓益肝颗粒的质量控制方法。
Objective: To establish a method for the determination of Astragaloside IV in QiLingYiGan granules. Methods: Quantitative analysis of Astragaloside IV in Qilinglinggan granule was carried out by high performance liquid chromatography coupled with evaporative light scattering detection (HPLC-ELSD). The column was Aglient Eclipse XDB C18 (4.6 mm × 250 mm, 5 μm) - water (35:65) as the mobile phase at a flow rate of 1.0 mL · min-1 at a column temperature of 25 ℃. The evaporative light scattering (ELSD) detector was used. The drift tube temperature was 110 ℃ and the carrier gas flow rate was 1.6 L · min-1. Injection volume 10μL. Results: Astragaloside showed a good linearity between 0.498 and 9.96 μg, r = 0.9998. The average recovery was 98.05% and the RSD was 0.89%. The stability was good within 8 h at room temperature. Conclusion: HPLC-ELSD method is simple, rapid, accurate, reproducible and reliable. It can be used as a quality control method for SQLYG.
