采用乙醇回流提取的方法筛选了从甜瓜蒂中提取葫芦素B的最佳工艺。方法:葫芦素B的含量为评价指标,用正交试验的方法对乙醇浓度、料液比、提取时间、提取次数4个因素进行了考察,采用高效液相色谱法测定葫芦素B的含量:色谱柱为XDB-C18(4.6mm×150mm),流动相为甲醇-水(60:40,v/v),流速为0.8mL/min,柱温25℃,检测波长为228nm。结果:得到最佳工艺条件为:以65%乙醇为溶剂,在80℃条件下,料液比11:1,回流提取0.5h,提取次数3次,其葫芦素B的相对提取率可达86.15%。结论:优选所得工艺合理可行。
The optimal extraction process of cucurbitacin B from melon was screened by ethanol reflux extraction. Methods: The content of cucurbitacin B was used as evaluation index. The orthogonal test was used to study the factors of ethanol concentration, solid - liquid ratio, extraction time and extraction times. The content of cucurbitacin B was determined by high performance liquid chromatography (HPLC) The column was XDB-C18 (4.6 mm × 150 mm). The mobile phase consisted of methanol-water (60:40, v / v) and the flow rate was 0.8 mL / min. The column temperature was 25 ℃ and the detection wavelength was 228 nm. Results: The optimum conditions were as follows: the relative extraction rate of cucurbitacin B was 86.15 with 65% ethanol as the solvent, under the condition of 80 ℃, the ratio of solid to liquid was 11: 1, %. Conclusion: The preferred process is reasonable and feasible.
