帕金森病是常见的中枢神经系统退行性疾病,以黑质和纹状体多巴胺能神经元进行性缺失及α-synuclein聚集和Lewy小体的形成为主要特点,其确切的病因及发病机制尚不清楚。近年发现蛋白质的构象发生改变可以导致一些所谓的蛋白质构象病,蛋白质结构改变与疾病的关系已成为分子生物学新的研究前沿,蛋白质构象发生改变在帕金森病发病中的作用越来越受到人们的关注。本文就帕金森病相关蛋白质构象发生改变机制及其之间的相互关系及对帕金森病发病过程的影响研究进展进行综述。
Parkinson’s disease is a common degenerative disease of the central nervous system. The main features are the progressive loss of dopaminergic neurons in substantia nigra and striatum, the accumulation of α-synuclein and the formation of Lewy bodies. The exact etiology and pathogenesis of Parkinson’s disease Not clear. In recent years, it has been found that the conformational changes of proteins can lead to some so-called protein conformation diseases. The relationship between protein structural changes and diseases has become a new frontier in molecular biology. The role of changes in protein conformation in the pathogenesis of Parkinson’s disease is getting more and more people s concern. This review summarizes the research progress on the mechanism of the alteration of the protein conformation related to Parkinson’s disease and its interrelationship and the influence on the pathogenesis of Parkinson’s disease.
