目的分析miR-18a和miR-328在肺腺癌A549细胞中的表达,并探讨下调miR-18a或miR-328的表达对其侵袭和迁移能力的影响。方法用荧光定量PCR检测A549细胞中miR-18a和miR-328的表达;通过转染miR-18a抑制物(miR-18a inhibitor)或miR-328 inhibitor,下调miR-18a或miR-328的表达,采用TranswellTM侵袭、迁移实验分析A549细胞侵袭和迁移能力的变化。结果 A549细胞中miR-18a和miR-328均呈高表达;而转染相应抑制物后miR-18a、miR-328表达下降,A549细胞的侵袭、迁移能力均明显降低。结论下调A549细胞miR-18a或miR-328水平可有效抑制肿瘤细胞的侵袭、迁移。
Objective To analyze the expression of miR-18a and miR-328 in lung adenocarcinoma A549 cells and to explore the effects of miR-18a or miR-328 expression on invasion and migration. Methods The expression of miR-18a and miR-328 in A549 cells was detected by real-time PCR. The expression of miR-18a or miR-328 was down-regulated by transfection with miR-18a inhibitor or miR- Transwell invasion and migration assays were used to analyze the invasion and migration of A549 cells. Results The expression of miR-18a and miR-328 were all highly expressed in A549 cells. However, the expression of miR-18a and miR-328 decreased after transfected with the corresponding inhibitor, and the invasion and migration of A549 cells were significantly decreased. Conclusion The down-regulation of miR-18a or miR-328 in A549 cells can effectively inhibit the invasion and migration of tumor cells.
