目的探讨Toll样受体4(TLR4)和Toll样受体9(TLR9)基因在正常宫颈组织、宫颈上皮内瘤变(CIN)组织和宫颈癌组织中的表达及意义。方法采用qRT-PCR法检测正常宫颈、CIN及宫颈癌各30例组织中TLR4和TLR9基因表达水平及其下游分子NF-κB和IFN-γ表达。分析TLR4和TLR9基因表达与NF-κB和IFN-γ表达的相关性。结果CIN、宫颈癌组织的TLR4、TLR9、NF-κB和IFN-γ表达水平均高于正常宫颈(P<0.01或P<0.05),且宫颈癌组织的TLR4、TLR9、NF-κB和IFN-γ表达水平均高于CIN组织(P<0.01)。TLR4与NF-κB、IFN-γ表达呈正相关(r=0.905、r=0.873,P<0.01),TLR9与NF-κB、IFN-γ表达亦呈正相关(r=0.953、r=0.976,P<0.01)。结论 TLR4和TLR9可能参与宫颈病变的恶性转变过程。
Objective To investigate the expression and significance of Toll-like receptor 4 (TLR4) and Toll-like receptor 9 (TLR9) gene in normal cervical tissue, cervical intraepithelial neoplasia (CIN) and cervical cancer. Methods The qRT-PCR was used to detect the expression of TLR4 and TLR9 and the expression of NF-κB and IFN-γ in 30 cases of normal cervix, CIN and cervical cancer. The correlation between TLR4 and TLR9 gene expression and the expression of NF-κB and IFN-γ was analyzed. Results The expressions of TLR4, TLR9, NF-κB and IFN-γ in CIN and cervical cancer tissues were significantly higher than those in normal cervix (P <0.01 or P <0.05), and the expressions of TLR4, TLR9, NF- γ expression levels were higher than CIN (P <0.01). The expression of TLR4 was positively correlated with the expression of NF-κB and IFN-γ (r = 0.905, r = 0.873, P <0.01) 0.01). Conclusion TLR4 and TLR9 may participate in the malignant transformation of cervical lesions.
