目的 :观察新型选择性磷酸二酯酶 V型强效抑制剂 E40 2 1,对顿抑心肌局部收缩功能的影响。方法 :闭塞麻醉开胸犬左冠状动脉前降枝 (L AD) 15 min伴 3 h再灌注 ,闭塞前 15 m in连续静脉给药 ,30 m in滴完 ;用心内膜下植入超声波微晶体法测局部心肌活动。结果 :E40 2 110μg· kg- 1 · min- 1 对心率 ,平均血压 ,二项乘积 ,左室最大上升速率 (L V dp/ dtmax)无明显经时影响 ,但使闭塞 L AD所致缺血区的心肌节段收缩率从 (17.3± 1.4) %减为 (1.0± 1.7) % ,再灌流后立即恢复 ,3 h后达到原来的 (11.1± 1.8) % ,并略高于溶媒组 (P <0 .0 5 )。结论 :E40 2 1在不影响实验性麻醉犬心脏血流动力学的剂量下 ,对顿抑心肌有改善或保护倾向。
OBJECTIVE: To observe the effect of E40 2 1, a novel and selective phosphodiesterase type V inhibitor, on the local contractile function of stunned myocardium. Methods: The left anterior descending artery (LAD) of occluded thoracotomy dogs was anesthetized for 15 min followed by 3 h of reperfusion. The animals were continuously injected intravenously 15 min before occlusion, Body method to measure local myocardial activity. Results: E40 2 110μg · kg-1 · min-1 had no obvious effect on heart rate, mean blood pressure, the product of two products and the maximum rate of left ventricular (LV dp / dtmax) (17.3 ± 1.4)% to (1.0 ± 1.7)%, resumed immediately after reperfusion, reached the original (11.1 ± 1.8)% after 3 h and slightly higher than that of vehicle group (P < 0 .0 5). CONCLUSION: E40 2 1 tends to improve or protect the stunned myocardium at a dose that does not affect the cardiohemodynamics of experimental anesthetized dogs.
