目的探讨多价肺炎链球菌荚膜多糖结合物对小鼠免疫应答的影响。方法分别用单一载体(破伤风类毒素)和混合载体(破伤风类毒素和白喉类毒素)作为多糖结合物的载体蛋白,制备5~9价肺炎链球菌荚膜多糖结合物,免疫NIH小鼠,采用间接ELISA法测定免疫小鼠血清中抗荚膜多糖特异性IgG抗体水平。结果多价肺炎链球菌荚膜多糖结合物的免疫效果优于相应的单价结合物;随着结合物配制价数的增高,小鼠的免疫应答能力趋于下降;多价结合物中使用两种载体能缓解此现象,但载体含量过高时,依然会抑制免疫应答。结论多价肺炎链球菌荚膜多糖结合物免疫小鼠后,随着载体剂量的增高,会出现载体诱导的表位抑制现象,使小鼠的免疫应答反应减弱。两种载体蛋白同时使用能缓解此现象。
Objective To investigate the effect of multivalent S. pneumoniae capsular polysaccharide conjugates on immune response in mice. Methods Single carrier (tetanus toxoid) and mixed carrier (tetanus toxoid and diphtheria toxoid) were used as carrier protein of polysaccharides conjugate, respectively, to produce 5-9-valent pneumococcal capsular polysaccharide conjugates, and to immunize NIH mice , The level of anti-capsular polysaccharide-specific IgG antibody in the serum of the immunized mice was determined by indirect ELISA. Results The immunogenicity of multivalent S. pneumoniae capsular polysaccharide conjugates was better than that of the corresponding monovalent conjugates. With the increase of the formulation valences of the conjugates, the immune response ability of mice tended to decrease. Two kinds of multivalent conjugates Vector can alleviate this phenomenon, but when the carrier is too high, it will still suppress the immune response. Conclusions After immunization of mice with multivalent S. pneumoniae capsular polysaccharide conjugate, the carrier-induced epitope suppression may occur with the increase of the carrier dosage, and the immune response of the mice will be weakened. The simultaneous use of two carrier proteins can alleviate this phenomenon.
