探讨FHIT基因与胃癌关系。方法 :采用PT -PCR及PCR产物直接测序法 ,对 2 6例胃癌组织及其配对正常组织的FHIT基因进行检测。结果 :7例 (2 6 .9% )胃癌组织存在异常FHIT转录本的表达 ,其中 1例无正常FHIT转录本的表达 ;另 6例同时存在正常FHIT转录本的表达 ;配对正常组织均无异常FHIT转录本的表达 ;测序证实异常转录本缺失 1个或 1个以上FHIT外显子。结论 :FHIT转录本的异常是胃癌中常见的和肿瘤特有的改变 ,FHIT基因的异常与我国部分胃癌的发生有关。
To investigate the relationship between FHIT gene and gastric cancer. METHODS: FHIT gene was detected in 26 gastric cancer tissues and their matched normal tissues by PT-PCR and direct sequencing of PCR products. Results: Abnormal expression of FHIT transcripts was found in 7 cases (26.9%) of gastric cancer tissues. One case had no expression of normal FHIT transcripts, and the other 6 cases had normal FHIT transcripts. There was no abnormality in paired normal tissues. Expression of FHIT transcripts; sequencing confirmed the absence of one or more FHIT exons in aberrant transcripts. Conclusion : The abnormality of FHIT transcript is a common and unique tumor change in gastric cancer. The abnormality of FHIT gene is related to the occurrence of some gastric cancer in China.
