目的探讨丙酮酸乙酯对滑膜成纤维细胞增殖的影响。方法收集胶原诱导的关节炎大鼠(collagen induced arthritis,CIA)和正常大鼠滑膜组织,培养滑膜成纤维细胞。光镜及免疫荧光法检测Vimentin蛋白鉴定滑膜成纤维细胞;分别用1、2、5、10、20 mmol/L浓度的丙酮酸乙酯作用于滑膜成纤维细胞;流式细胞术及CCK8法检测细胞增殖情况;流式细胞术检测细胞凋亡情况。结果显微镜观察细胞形态特征符合滑膜成纤维细胞,且Vimentin蛋白表达阳性;5种剂量的丙酮酸乙酯均能抑制细胞增殖同时诱导细胞凋亡,但是未见明显剂量依赖效应,以5 mmol/L剂量组作用效果最为明显。结论丙酮酸乙酯能够抑制滑膜成纤维细胞增殖同时促进其凋亡,且以5 mmol/L剂量组作用最为明显。
Objective To investigate the effect of ethyl pyruvate on the proliferation of synovial fibroblasts. Methods Collagen-induced arthritis (CIA) and synovial tissue of normal rats were collected and synovial fibroblasts were cultured. The vimentin protein was identified by light microscopy and immunofluorescence staining for synovial fibroblasts. The cells were treated with 1, 2, 5, 10 and 20 mmol / L ethyl pyruvate respectively. The results of flow cytometry and CCK8 France to detect cell proliferation; flow cytometry to detect apoptosis. Results The morphology of the cells was found to be synovial fibroblasts and the expression of Vimentin was observed by microscope. Ethyl pyruvate at 5 doses could inhibit cell proliferation and induce apoptosis at the same time. However, there was no significant dose - dependent effect. 5 mmol / L dose group the most obvious effect. Conclusion Ethyl pyruvate can inhibit the proliferation of synovial fibroblasts and promote their apoptosis at the dose of 5 mmol / L.
