目的探讨摄入贵州某54°白酒后,不同时程及不同剂量对大鼠睾丸间质细胞内类固醇激素急性调节蛋白(St AR)表达及血清睾酮水平的变化,以期为科学饮酒提供基础资料。方法正常成年雄性SD大鼠69只,随机分为正常对照组(n=15)及实验组(n=54)。实验组分为低剂量组0.8ml/(kg·d)、中剂量组1.6ml/(kg·d)及高剂量组2.4ml/(kg·d),实验组大鼠每日灌胃2次。分别于第4周末、第8周末、第12周末采血清,取睾丸组织。采用免疫组织化学SABC法、图像分析、Western blotting检测睾丸组织St AR的表达,采用化学发光法检测大鼠血清睾酮水平。结果各实验组的血清睾酮水平与正常对照组比较未见明显异常,差异无统计学意义(P>0.05)。St AR免疫组织化学阳性产物存在于睾丸间质细胞胞质内;与正常对照组比较,4周、8周、12周各剂量组St AR免疫染色及平均吸光度均有所增强(P<0.05);Western blotting检测,4周、8周、12周各剂量组St AR蛋白表达水平增强(P<0.05)。结论在本实验设定的剂量和时程内用该白酒灌胃后,大鼠睾丸间质细胞内St AR表达水平有所增强,血清睾酮水平并无明显变化。
Objective To investigate the changes of St AR expression and serum testosterone levels in testis stromal cells of different degrees and different doses of 54 ° liquor in Guizhou in order to provide basic data for scientific drinking. Methods Sixty-nine adult male Sprague-Dawley rats were randomly divided into normal control group (n = 15) and experimental group (n = 54). The experimental groups were 0.8ml / (kg · d) in the low dose group, 1.6ml / (kg · d) in the middle dose group and 2.4ml / (kg · d) in the high dose group. . At the end of the 4th, 8th and 12th week respectively, serum was collected and testicular tissue was taken. Immunohistochemistry SABC method, image analysis and Western blotting were used to detect the expression of St AR in testis tissue. The level of serum testosterone was detected by chemiluminescence. Results There was no significant difference between the two groups in serum testosterone levels and normal controls (P> 0.05). St AR immunohistochemical positive products were found in the cytoplasm of testicular stromal cells. St AR immunostaining and average absorbance of each dose group at 4 weeks, 8 weeks and 12 weeks were significantly increased (P <0.05) The protein expression of St AR in each dose group increased at 4 weeks, 8 weeks and 12 weeks (P <0.05) by Western blotting. Conclusions The intragastric administration of the liquor within the dose and schedule set in this experiment enhanced the expression of St AR in rat leydig cells and did not change the level of serum testosterone.
