目的了解2013-2015年漳州市麻疹病毒基因型流行情况。方法用Real-time RT-PCR法对2013-2015年漳州市的392份疑似麻疹病例咽拭子标本进行核酸检测;用RT-PCR法扩增核酸阳性标本的N基因羧基末端594个核苷酸片段,并对扩增产物测序;以N基因羧基末端450个核苷酸序列构建系统进化树,并进行核苷酸和氨基酸同源性分析。结果 28份标本麻疹病毒核酸阳性,阳性率7.1%(28/392);获得21份麻疹病毒的核苷酸序列,经基因型鉴定18株为H1a基因亚型,1株为A基因型,2株为D8基因型。漳州市所有的H1a亚型麻疹病毒与H1a亚型参考株China93-2-AF045205.1-H1a之间核苷酸和氨基酸同源性分别为98.0%-99.1%和97.3%-99.3%。A基因型麻疹病毒与疫苗株Shanghai-191-DQ390225-A的核苷酸和氨基酸同源性分别为99.6%和99.3%。2株D8基因型麻疹病毒之间核苷酸和氨基酸同源性均为100%,它们与世界卫生组织(WHO)参考株MVi/Manchester.GBR/30.94-AF280803-D8的核苷酸和氨基酸同源性分别为96.4%和96.7%。结论 2013-2015年漳州市麻疹病毒优势基因型为H1a基因亚型;A基因型为疫苗相关株;漳州市首次出现D8基因型麻疹病毒输入性病例。
Objective To understand the prevalence of measles virus in Zhangzhou City from 2013 to 2015. Methods Real-time RT-PCR was used to detect the nucleic acid of 392 throat swab specimens in suspected measles cases in Zhangzhou City from 2013 to 2015. The 594 nucleotides of N-terminal carboxyl end of nucleic acid positive samples were amplified by RT-PCR The sequence of the amplified product was sequenced. The phylogenetic tree was constructed based on the 450 nucleotide sequences of the carboxyl terminus of N gene and nucleotide and amino acid homology analysis. Results 28 samples were positive for measles virus, the positive rate was 7.1% (28/392). The nucleotide sequences of 21 measles viruses were obtained. Among them, 18 were H1a genotypes, 1 was genotype A, 2 Strain was D8 genotype. The nucleotide and amino acid homology of all H1a subtype measles virus and H1a subtype reference strain China93-2-AF045205.1-H1a in Zhangzhou were 98.0% -99.1% and 97.3% -99.3%, respectively. The nucleotide and amino acid homologies of the A genotype measles virus and the vaccine strain Shanghai-191-DQ390225-A were 99.6% and 99.3%, respectively. The nucleotide and amino acid homologies between the two D8 genotypes were all 100%. They were identical to those of the reference strain MVi / Manchester.GBR / 30.94-AF280803-D8 of the World Health Organization (WHO) The source was 96.4% and 96.7% respectively. Conclusion The predominant genotypes of measles virus in Zhangzhou City from 2013 to 2015 were H1a genotypes, A genotypes were vaccine-associated strains, and the first case of D8 genotype was reported in Zhangzhou City.
