目的验证丙烯酸-2-乙基己酯(2-EHA)的遗传毒性。方法采用鼠伤寒沙门菌回复突变试验(Ames试验),小鼠骨髓嗜多染红细胞微核试验和小鼠淋巴瘤细胞TK基因突变试。Ames试验设每皿0.08、0.4、2.0、10.0和50.0μl剂量组,另设自发对照组、溶剂对照组和阳性对照组;微核试验设278、555、1 110和2 220 mg/kg·bw剂量组,另设溶剂对照和阳性对照组;TK基因突变试验设12.5、25、50和100μmol/L剂量组,另设溶剂对照和阳性对照组。结果 Ames试验中,加或不加S9的情况下,受试物对TA97、TA98、TA100和TA102菌株均未显示致突变作用。微核试验中,各剂量组微核率与对照组比较,差异无统计学意义。TK基因突变试验两较高剂量组突变频率均高于溶剂对照100×10-6以上,试验结果为阳性。结论综合考虑前期试验结果及国外相关研究,认为丙烯酸-2-乙基己酯具有遗传毒性的可能性较小。
Objective To verify the genotoxicity of 2-ethylhexyl acrylate (2-EHA). Methods Salmonella typhimurium back-mutagenesis test (Ames test), mouse bone marrow polychromatic erythrocyte micronucleus test and mouse lymphoma cell TK gene mutation test. Ames test set 0.08,0.4,2.0,10.0 and 50.0μl per dish dose group, another set of spontaneous control group, solvent control group and positive control group; micronucleus test set 278,555,1 110 and 2 220 mg / kg · bw Dose group, and another solvent control and positive control group; TK gene mutation test set 12.5, 25, 50 and 100μmol / L dose group, another set of solvent control and positive control group. Results Ames test, with or without S9 case, the test substance on TA97, TA98, TA100 and TA102 strains did not show mutagenic effect. In the micronucleus test, there was no significant difference in the micronucleus rate of each dose group compared with the control group. Mutation frequency of TK gene mutation test in the two higher dose groups were higher than the solvent control 100 × 10-6 above, the test result was positive. Conclusion Considering the results of previous studies and related studies abroad, it is concluded that 2-ethylhexyl acrylate is less likely to be genotoxic.
