目的:以高效液相色谱法测定鸡、灰鸡和黄鸡3种鸡菌子实体中核苷及嘌呤碱基类成分的组成和含量,建立鸡类真菌核苷及嘌呤碱基类成分的控制指标。方法:采用Waters Xbridge C_(18)柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸水溶液为流动相,流速为0.8 mL·min~(-1),柱温为30℃,检测波长为260 nm。结果:3种鸡菌子实体中均检出次黄嘌呤、黄嘌呤、胞苷、腺苷、鸟苷、尿苷、肌苷等成分,且在质量浓度分别在7.51~120.12、4.99~79.84、25.13~402.00、12.61~201.80、5.03~80.48、5.00~80.00、10.00~160.00μg·mL~(-1)与峰面积呈良好线性关系(r≥0.999 6);平均回收率为95.83%~103.01%,RSD≤3.98%(n=6)。鸡中2种嘌呤碱基、5种核苷类成分含量分别为:0.36、3.13、2.20、3.63、1.11、1.87、1.50 mg·g~(-1);灰鸡含量为:0.10、0.78、1.86、3.07、1.48、1.70、0.62 mg·g~(-1);黄鸡含量为:0.22、2.53、1.86、2.35、0.78、0.84、0.83 mg·g~(-1)。结论:本实验建立的HPLC测定鸡类真菌中5种核苷及2种嘌呤碱基类成分含量的分析方法简单、重复性好、准确度高,可以为鸡类真菌的质量控制提供参考,3种鸡菌子实体在医疗和营养保健上都有较高的研究价值。
OBJECTIVE: To determine the composition and content of nucleoside and purine base components in three kinds of Agrocybe aegeritus from chicken, sea cucumber, and yellow chicken by high performance liquid chromatography (HPLC) Control of components of the class. Methods: A Waters Xbridge C 18 column (250 mm × 4.6 mm, 5 μm) was used with a mobile phase of methanol-0.1% phosphoric acid at a flow rate of 0.8 mL · min -1 and a column temperature of 30 ℃. The wavelength is 260 nm. Results: The constituents of hypoxanthine, xanthine, cytidine, adenosine, guanosine, uridine, inosine and so on were detected in all the three kinds of Agrobacterium tumefaciens fruiting bodies, and the mass concentrations were 7.51-120.12,4.99-79.84, 25.13 ~ 402.00,12.61 ~ 201.80,5.03 ~ 80.48,5.00 ~ 80.00,10.00 ~ 160.00μg · mL ~ (-1) and the peak area showed a good linear relationship (r≥0.9996); the average recoveries ranged from 95.83% to 103.01% , RSD≤3.98% (n = 6). Gynostemma in two kinds of purine base, five kinds of nucleosides content were: 0.36,3.13,2.20,3.63,1.11,1.87,1.50 mg · g -1; gray chicken gall content: 0.10,0.78 , 1.86,3.07,1.48,1.70,0.62 mg · g -1, respectively. The content of yellow chicken was 0.22, 2.53, 1.86, 2.35, 0.78, 0.84 and 0.83 mg · g -1, respectively. CONCLUSION: The method for determination of five nucleosides and two kinds of purine bases in chicken galls by HPLC in this experiment is simple, reproducible and accurate, which can provide reference for the quality control of chicken galls. , The three kinds of Agrobacterium fruiting bodies have higher research value in medical treatment and nutrition and health care.
