目的研究吗啡预先给药对急性心肌缺血大鼠心肌NF-κB水平的影响,探讨吗啡的心肌保护作用机制。方法健康雄性SD大鼠54只,随机分为3组(n=18):假手术组(S组)、心肌缺血组(I组)、吗啡组(M组)。I、M组开胸后结扎冠状动脉左前降支,S组仅穿线不结扎。M组在结扎冠状动脉15 min前经尾静脉注射吗啡1.25 mg·kg-1,S、I组注射等量生理盐水。缺血6 h后处死动物,取缺血区心肌组织观察病理学改变,应用免疫组织化学法检测心肌NF-κB表达,用流式细胞术检测心肌NF-κB激活情况。结果光镜下M组心肌缺血情况比I组重,但比S组轻;I组心肌NF-κB表达及激活百分率大于M组,但小于S组(P<0.05)。结论吗啡预先给药在一定程度上抑制心肌缺血大鼠心肌NF-κB的表达、激活,对心肌有一定的保护作用。
Objective To investigate the effect of morphine preconditioning on myocardial NF-κB in rats with acute myocardial ischemia and to explore the mechanism of myocardial protection of morphine. Methods Fifty - four healthy male Sprague Dawley rats were randomly divided into 3 groups (n = 18): sham operation group (S group), myocardial ischemia group (I group) and morphine group (M group). In the I and M groups, the left anterior descending coronary artery was ligated after thoracotomy. In the S group, only the ligature was not ligated. In group M, morphine 1.25 mg · kg-1 was injected into caudal vein 15 min prior to ligation of coronary artery. Rats in group S and I were injected with the same amount of saline. Animals were sacrificed at 6 hours after ischemia. Pathological changes were observed in ischemic myocardium. The expression of NF-κB was detected by immunohistochemistry. The activation of NF-κB was detected by flow cytometry. Results Myocardial ischemia was more severe in group M than in group I, but less in group M than in group S. The expression and activation of NF-κB in group I were higher than those in group M, but less than those in group S (P <0.05). Conclusion Pretreatment with morphine can inhibit the expression and activation of NF-κB in myocardium to a certain extent.
