将补肾中药按水煎浓缩、水煎醇沉和乙醇提取三种传统加工工艺制成口服液,观察它们对氯化铝溶液所致拟痴呆小鼠记忆功能的影响。每种药液均以高(10mL/k/日)和低(3.3mL/kg/日)两种剂量灌胃,并以同龄、同种小鼠为对照组。在喂药过程中,每10天用避暗法测定各组每只小鼠记忆功能。连续喂药30天后,断头分离出每只小鼠的大脑皮层组织,用酶底物反应和受伤协郎配某结会分析.测字各组小鼠大脑皮层组织中乙酞胆碱酯酶(A。*E)活性和胆碱能M受体结合容量(Rt)值。实验结果表明,拟痴呆小鼠大脑皮层组织中A。hE活性较对照组明显升高(P<0.05),M受体Rt值显著下降(P<0.05)。三种不同加工工艺药液的高低两种剂量都能明显降低A。hE活性,升高M受体Rt值,与同组治疗前比较,差异显著(P<0.05—0.001)。与小鼠整体迷宫实验能显著提高记忆力相一致。各组间比较差异不明显。证明三种不同制药工艺都可以提取补肾中药的有效成分。水煎浓缩方法简便,价格便宜,应为首选方法。
The kidney traditional Chinese medicines were made into oral liquids by decocting, decoction, alcohol precipitation and ethanol extraction, and their effects on the memory function of dementia mice induced by aluminum chloride solution were observed. Each drug solution was intragastrically administered at high (10 mL/k/day) and low (3.3 mL/kg/day) doses, and the same age and same-type mice were used as control groups. During the course of feeding, the memory function of each mouse in each group was measured by a dark-separation method every 10 days. After 30 days of continuous drug administration, cerebral cortex tissue was isolated from each mouse and analyzed by enzyme substrate reaction and injury. The activity of cholinesterase (A.*E) and cholinergic M receptor binding capacity (Rt) were measured in the cerebral cortex of mice in each group. The experimental results showed that the dementia mice had cerebral cortex A. The activity of hE was significantly higher than that of the control group (P<0.05), and the M receptor Rt value was significantly decreased (P<0.05). Both high and low doses of three different processing methods can significantly reduce A. The hE activity increased the M receptor Rt value compared with the same group before treatment, the difference was significant (P<0.05-0.001). The overall maze experiment with mice can significantly improve memory. There was no significant difference between the groups. It proves that three different pharmaceutical processes can extract the active ingredients of kidney-invigorating Chinese medicine. The decocting and concentrating method is simple and inexpensive and should be the preferred method.
