目的:观察益气健脾方对CAG大鼠血清中Reg I和PCNA表达的影响及病理组织学的变化,探讨益气健脾方对CAG大鼠成模的抑制作用及其可能机制。方法:将60只SD大鼠随机分为正常组、模型组、益气健脾方低、中、高剂量组和阳性对照组,每组10只,采用MNNG联合饥饱失常的综合造模方法制备慢性萎缩性胃炎大鼠模型,10周后观察益气健脾方对CAG大鼠模型成模的影响,并测定血清中Reg I及PCNA表达含量等的变化情况。结果:1益气健脾方不同剂量组和阳性对照组对CAG大鼠在一般情况和病理状态等方面均有改善作用。2益气健脾方各组与阳性对照组对血清中Reg I及PCNA含量具有下调作用,与模型组相比具有显著差异(P<0.05),且益气健脾方高剂量组优于阳性对照组(P<0.05),呈现出一定的剂量依赖趋势。结论:益气健脾方对CAG大鼠成模具有抑制作用,能够改善CAG大鼠胃黏膜病理萎缩状态,其机理除与传统中医功效理论和药物组成的药理作用相关,与调控血清中Reg I及PCNA表达亦相关,这可能是其发挥效应的作用机制之一。
Objective: To observe the effect of Yiqi Jianpi Prescription on the expression of Reg I and PCNA and the histopathological changes of CAG rat serum, and to explore the inhibitory effect of Yiqi Jianpi Prescription on CAG rat model and its possible mechanism. Methods: Sixty Sprague-Dawley rats were randomly divided into normal group, model group, low, medium and high dose Yiqi Jianpi group and positive control group, with 10 rats in each group. The rat model of chronic atrophic gastritis was prepared. After 10 weeks, the effect of Yiqi Jianpi Prescription on the model of CAG rat model was observed. The content of Reg I and PCNA in serum were determined. Results: 1 Yiqi Jianpi Fang different dose groups and positive control group on CAG rats in general and pathological conditions have improved. There were significant differences between the two groups (P <0.05) and the high-dose Yiqi Jianpi Fang group Control group (P <0.05), showing a dose-dependent trend. Conclusion: Yiqi Jianpi Prescription can inhibit the formation of mold in CAG rats and ameliorate the pathological atrophy of gastric mucosa in CAG rats. The mechanism is related to the traditional Chinese medicine efficacy theory and the pharmacological effects of drug composition, And PCNA expression is also related, which may be one of the mechanisms of its effect.
