利用聚丙烯酰胺凝胶等电聚焦电泳法研究了甘蓝(BrasicaOleraceaL.)自交不亲和系与亲和系的柱头和花粉蛋白质谱带,并进行自交,以考察其亲和指数。结果表明,自交不亲和系与亲和系的柱头蛋白质谱带有明显差异:6条带只出现于自交不亲和系中;亲和系柱头没有特异带。对比自交不亲和系开花前2~3天的柱头与成熟柱头的蛋白质谱带,发现了S特异蛋白质的准确位置,其等电点约为8.3~8.5。此法可用于鉴定甘蓝自交不亲和性。以双蒸水和疏基乙醇作为提取液处理花粉,均未发现自交不亲和系的特征谱带。
The stigma and pollen protein bands of self-incompatible and inbred lines of Brassica oleracea L. were studied by polyacrylamide gel isoelectric focusing electrophoresis and self-cross-pollination to investigate their affinity index. The results showed that the stigma protein bands of self-incompatibility and affinity lines were significantly different: 6 bands appeared only in self-incompatible lines; Compared with the protein bands of stigma and mature stigma 2 to 3 days before flowering in the self-incompatible lines, we found the exact position of the S-specific protein with an isoelectric point of about 8.3-8.5. This method can be used to identify kale self-incompatibility. Double distilled water and spiky ethanol were used as the extracts to treat pollen, and no characteristic bands of self-incompatible lines were found.
