选择在合成PAI-1方面与肝细胞相似的肝胚胎瘤细胞株(HEPG2),用不同浓度的胰岛素、胰岛素原在不同时间内刺激单层HEPG2细胞,测培液中PAI—1活性。结果发现:胰岛素和胰岛素原均能刺激PAI-1的分泌,具有时间和剂量依赖性。不同浓度的胰岛素、胰岛素原在刺激8h内PAI-1的分泌差异性不明显,8h后超生理量的胰岛素、胰岛素原的刺激作用明显高于生理浓度的胰岛素和胰岛素原。
The HEPG2 cell line, which is similar to the hepatocyte in the synthesis of PAI-1, was selected to stimulate monolayer HEPG2 cells with different concentrations of insulin and proinsulin at different times to measure PAI-1 activity in the culture solution. The results showed that both insulin and proinsulin can stimulate the secretion of PAI-1 in a time- and dose-dependent manner. Different concentrations of insulin and proinsulin had no significant differences in the secretion of PAI-1 within 8 h after stimulation. After 8 h, the hyperphysiological amounts of insulin and proinsulin stimulated significantly more than physiological concentrations of insulin and proinsulin.
