目的:建立适合于瓜蒌种子的蛋白质提取方法。方法:比较水溶液直接提取法(3种)、三氯乙酸(TCA)-丙酮法、三羟甲基氨基甲烷(Tris)-丙酮法、蛋白质提取液提取法等6种蛋白质提取方法所获瓜蒌种子总蛋白含量、聚丙烯酰胺凝胶电泳的谱带等方面的差异。结果:蛋白质提取液提取法、TCA-丙酮法提取蛋白含量较高,但杂质多,电泳谱带严重展宽而且明显拖尾;Tris-丙酮法提出的蛋白质种类缺失严重;直接提取法提取蛋白含量少,但杂质少,电泳谱带清晰,离心2次谱带更加清晰。结论:水溶液直接提取法最适合用于聚丙烯酰胺凝胶电泳,蛋白质提取液提取法提出蛋白质含量最多。
Objective: To establish a method of protein extraction suitable for melon seeds. Methods: Six kinds of protein extraction methods such as direct extraction of aqueous solution (3 kinds), trichloroacetic acid (TCA) - acetone method, Tris - acetone method and protein extraction method were compared. Total protein content of seed, polyacrylamide gel electrophoresis band and so on. Results: Protein extraction method, TCA-acetone extraction of protein content is high, but more impurities, electrophoresis band with severe broadening and significant tailing; Tris-acetone method proposed serious lack of protein species; direct extraction method to extract protein content less , But less impurities, electrophoresis band clear, centrifugal 2 times the band more clearly. Conclusion: The direct extraction method of aqueous solution is the most suitable for polyacrylamide gel electrophoresis. Protein extraction method proposed the most protein content.
