以精品肝素为原料,研究超声波结合过氧化氢(H_2O_2)制备低分子肝素(2~8 ku)的工艺条件。以超声时间、超声功率、H_2O_2含量和超声温度为因素,采用单因素试验和响应面试验设计,以2~8 ku分子质量部分所占比例为测定指标,确定低分子肝素制备工艺参数的最优组合。研究结果表明,在超声时间76 min、超声功率405W、H_2O_2含量5.8%、超声温度61℃条件下,得到低分子肝素的平均分子质量(3900±50)u,2~8 ku组分含量的平均值为87.35%,低分子肝素的抗-FXa效价为101.16 IU/mg,抗-FIIa效价为31.13 IU/mg,抗-FXa/抗-FIIa值为3.25。
Using high-quality heparin as raw material, the technological conditions of preparing low-molecular-weight heparin (2-8 ku) by ultrasonic combined with hydrogen peroxide (H 2 O 2) were studied. Taking ultrasonic time, ultrasonic power, H 2 O 2 content and ultrasonic temperature as the factors, single factor experiment and response surface test design were used to determine the optimal preparation parameters of low molecular weight heparin with the proportion of 2 ~ 8 ku molecular weight fraction combination. The results showed that average molecular weight (3900 ± 50) u and average content of 2 ~ 8 ku of low molecular weight heparin were obtained under the condition of ultrasonic time of 76 min, ultrasonic power of 405W, H 2 O 2 content of 5.8% and ultrasonic temperature of 61 ℃ Value of 87.35%, low molecular weight heparin anti-FXa titer of 101.16 IU / mg, anti-FIIa titers of 31.13 IU / mg, anti-FXa / anti-FIIa value of 3.25.
