目的研究扶正剔邪搜络方对肺纤维化大鼠不同时相肺功能的影响。方法将128只SD大鼠随机分为假手术组、模型组、扶正剔邪搜络组、泼尼松组,每组32只。气管内注射博莱霉素(0.25ml/100g)建立肺纤维化模型,假手术组注射等体积生理盐水。造模24h后,扶正剔邪搜络组给予扶正剔邪搜络方0.74g/(kg·d),泼尼松组给予5mg/(kg·d),假手术组和模型组给予等体积生理盐水。给药后第7、14、28、45天,观察各组大鼠肺组织病理结果及吸气阻力(Ri)、肺顺应性(Cl)、用力肺活量(FVC)、第0.2秒末呼出容积(FEV0.2)、呼出气量为25%~75%肺活量时平均流量(FEF25-75%)变化。结果模型组第7天肺间质有不同程度炎症,第14天肺间质胶原纤维明显增多,第28天肺纤维区较14天增大、增多,第45天现“蜂窝肺”改变。扶正剔邪搜络组和泼尼松组肺泡炎与纤维化程度均轻于模型组。与模型组同时间比较,扶正剔邪搜络组第14天时Cl、FVC均明显升高(P<0.01),第28天时Ri、Cl、FVC、FEV0.2、FEF25-75%差异有统计学意义(P<0.01),第45天时Ri、Cl、FEV0.2、FEF25-75%差异有统计学意义(P<0.05或P<0.01)。与泼尼松组同时间比较,扶正剔邪搜络组45天时Ri明显降低(P<0.01),CI明显升高(P<0.01)。结论扶正剔邪搜络方可改善肺纤维化大鼠肺功能,早期介入并维持一定疗程作用最明显。
Objective To study the influence of Fuzheng Decoction on the lung function in different phases of pulmonary fibrosis in rats. Methods 128 SD rats were randomly divided into sham operation group, model group, Fuzheng Decoction and Prednisone group, with 32 rats in each group. Intratracheal injection of bleomycin (0.25ml / 100g) model of pulmonary fibrosis, sham operation group injected with equal volume of saline. After modeling for 24 hours, the rats in Fuzheng Decoction group were given 0.74 g / (kg · d) Fuzheng Decoction, and 5 mg / (kg · d) in the prednisone group. The rats in sham operation group and model group were given equal volume of physiology brine. On the 7th, 14th, 28th and 45th day after the administration, the pathological results of the lung tissues were observed and the inspiratory resistance (Ri), pulmonary compliance (Cl), forced vital capacity (FVC) FEV0.2), exhaled volume of 25% to 75% of the average volume flow (FEF25-75%) changes. Results At the 7th day in the model group, interstitial lung tissue had different degrees of inflammation. On the 14th day, the interstitial collagen fibers increased significantly. On the 28th day, the lung fiber area increased and increased compared with that on the 14th day. On the 45th day, the “cellular lung” changed . Fuzheng tick evil search group and prednisone alveolitis and fibrosis were lighter than the model group. Compared with the model group at the same time, Cl and FVC were significantly increased on the 14th day in Fuzhengxuexueluo group (P <0.01). On the 28th day, the differences of Ri, Cl, FVC, FEV0.2 and FEF25-75% were statistically significant (P <0.01). The difference of Ri, Cl, FEV0.2 and FEF25-75% on day 45 was statistically significant (P <0.05 or P <0.01). Compared with the prednisone group, Ri significantly decreased (P <0.01) and CI significantly (P <0.01) on the 45th day in Fuzheng Decoction. Conclusion Fuzheng tick evil Search and Search can improve lung function in rats with pulmonary fibrosis, early intervention and maintain a certain course of treatment the most obvious.
