目的:研究3种来自大籽獐牙菜的酮类化合物1,8-二羟基-3,7-二甲氧基酮(methylswertianin,MET)、1,5,8-三羟基-3-甲氧基酮(bellidifodin,BEL)和1-羟基-3,7,8-三甲氧基酮(decussatin,DEC)的抗氧化作用。方法:采用H2O2和VC-Fe2+两种羟自由基发生系统;硫代巴比妥酸法测定大鼠肝匀浆及肝线粒体中脂质过氧化物丙二醛(MDA)含量;分光光度法测定大鼠红细胞氧化溶血和肝线粒体膨胀程度研究酮的抗氧化效果。结果:3种酮均能有效抑制MDA的产生,减少大鼠红细胞氧化性溶血,减轻肝线粒体膨胀程度;其中BEL效果最好,其次是MET和DEC。结论:酮具有明显的抗脂质过氧化作用。
OBJECTIVE: To study three kinds of steroidal compounds, 1,8-dihydroxy-3,7-dimethoxyanthrone (MET), 1,5,8-trihydroxy-3-one from Swertia macrocarpa. Antioxidation of bellidifodin (BEL) and 1-hydroxy-3,7,8-trimethoxydecane (DEC). METHODS: H2O2 and VC-Fe2+ two hydroxyl radical generating systems were used; thiobarbituric acid method was used to determine the content of lipid peroxide malondialdehyde (MDA) in rat liver homogenate and liver mitochondria; spectrophotometric determination Study on Oxidative Hemolysis and Mitochondrial Swelling of Rat Erythrocytes and Its Antioxidative Effect . RESULTS: All three kinds of indolinone could effectively inhibit the production of MDA, reduce red blood cell oxidative hemolysis and reduce liver mitochondrial swelling. Among them, BEL had the best effect, followed by MET and DEC. Conclusion: Anthrone has obvious anti-lipid peroxidation effect.
