目的:探讨肝细胞癌(hepatocellular carcinoma,HCC)组织内及体外细胞培养中磷脂酰肌醇蛋白聚糖-3(glypican-3,GPC3)与细胞增殖的关系.方法:采用免疫组织化学方法检测54例HCC组织中GPC3的表达,并对比其与Ki-67标记指数及各种临床病理参数的关系.使用shRNA抑制SMMC7721肝癌细胞系中GPC3的表达,MTT检测肝癌细胞的增殖变化.结果:在HCC组织中,GPC3与Ki-67指数有关(r=0.521,P=0.000),也与分化程度、血管浸润情况具有相关性(r=0.287,0.289;P=0.035,0.034).体外实验结果显示,抑制GPC3表达后,实验组细胞增殖活性明显低于阴性对照组和空白对照组(F=45.38,P=0.000).结论:GPC3在HCC的发生、发展和浸润过程中起重要作用,主要与影响肝癌细胞增殖有关.
Objective: To investigate the relationship between glypican-3 (GPC3) and cell proliferation in hepatocellular carcinoma (HCC) tissue and in vitro.Methods: Immunohistochemistry was used to detect the expression of glypican-3 The expression of GPC3 in HCC tissues was compared with the Ki-67 labeling index and various clinicopathological parameters. The expression of GPC3 in SMMC7721 hepatoma cell line was inhibited by shRNA, and the proliferation of hepatocellular carcinoma cells was detected by MTT assay.Results: GPC3 was associated with Ki-67 index (r = 0.521, P = 0.000), but also with the degree of differentiation and vascular infiltration (r = 0.287,0.289; P = 0.035,0.034) .In vitro, The inhibition of GPC3 expression in experimental group was significantly lower than that in negative control group and blank control group (F = 45.38, P = 0.000) .Conclusion: GPC3 plays an important role in the occurrence, development and invasion of HCC, Liver cancer cell proliferation related.
