小分子量细胞因子的~(125)I同位素标记是研究细胞因子作用机理及其检测十分有用的技术。由于细胞因子分子量通常较小,~(125)I的直接偶连常导致生物活性丧失。我们在~(125)I标记重组人γ-干扰素的实验中,比较了几种连接方法后,用自制的Bolton-Hunter酰化试剂标记取得了较满意的结果。现报道如下。
The ~ (125) I isotope labeling of small molecular weight cytokines is a very useful technique to study the mechanism of action of cytokines and its detection. Due to the generally small molecular weight of cytokines, the direct coupling of ~ (125) I often leads to the loss of biological activity. In the experiment of ~ (125) I labeled recombinant human interferon-gamma, we compared the connection methods and obtained the satisfactory results with the homemade Bolton-Hunter acylation reagent.
