目的:研究左旋棉酚对人前列腺癌PC-3细胞体外增殖和凋亡的影响。方法:MTT法检测左旋棉酚对PC-3细胞增殖的影响,透射电镜观察细胞超微结构的改变,TUNEL染色观察细胞凋亡,流式细胞术检测细胞周期的改变,半定量RT-PCR检测Bcl-2及Bak mRNA的表达水平。结果:左旋棉酚能显著抑制PC-3细胞的体外增殖,并呈浓度-时间依赖性。左旋棉酚可以诱导PC-3细胞发生典型的凋亡形态学改变和G0/G1期阻滞,RT-PCR显示Bcl-2 mRNA表达水平降低,Bak mRNA表达水平升高。结论:左旋棉酚能诱导前列腺癌细胞的体外凋亡,主要可能与Bak mRNA的表达升高及Bcl-2 mRNA的表达下调有关。
Objective: To investigate the effect of L-gossypol on the proliferation and apoptosis of human prostate cancer PC-3 cells in vitro. METHODS: The effects of L-Gossypol on the proliferation of PC-3 cells were detected by MTT assay. The ultrastructural changes of cells were observed by transmission electron microscope. Cell apoptosis was observed by TUNEL staining. Cell cycle was detected by flow cytometry. Semiquantitative RT-PCR Bcl-2 and Bak mRNA expression levels. RESULTS: L-Gossypol significantly inhibited the proliferation of PC-3 cells in vitro and in a time-and concentration-dependent manner. L-gossypol can induce typical morphological changes of apoptosis and G0 / G1 arrest in PC-3 cells. The mRNA expression of Bcl-2 was decreased and the expression of Bak mRNA was increased by RT-PCR. CONCLUSION: L-Gossypol can induce apoptosis of prostate cancer cells in vitro, which may be related to the increase of Bak mRNA expression and the down-regulation of Bcl-2 mRNA expression.
