目的探索一种观察细胞形态结构的新的实验技术和方法。了解Hep-2细胞的形态结构特点。方法 取培养48 小时后的Hep-2细胞(培养的人喉鳞癌细胞株),直接滴片法将细胞悬液滴在PMMA上,以同步辐射装置产生的软X射线作光源进行SXCM成像研究。结果SXCM技术观察HeP-2细胞可清楚地显现细胞膜和核膜表面有很多皱褶,凹凸不平,而光镜下无法看清这种结构特点。结论SXCM技术在观察活细胞生理状态下的形态结构时具有比光镜更独特的优点。它不需要脱水、固定、染色等一系列准备工作。
Objective To explore a new experimental technique and method for observing the morphology of cells. Learn Hep-2 cells morphological characteristics. Methods Hep-2 cells (cultured human laryngeal squamous carcinoma cell line) cultured for 48 hours were directly dripped onto PMMA by droplet method. SXCM imaging was performed with soft X-ray generated by the synchrotron radiation device as a light source . Results SXCM technology observed HeP-2 cells can clearly show the cell membrane and nuclear membrane surface has a lot of folds, uneven, and light microscope can not see this structural features. Conclusion SXCM has more advantages than light microscopy in observing the morphological structure of living cells under physiological conditions. It does not require dehydration, fixing, dyeing and a series of preparatory work.
