目的优化测定癫痫患儿血清中丙戊酸钠浓度的方法。方法以环己烷羧酸作为内标,血样经乙腈蛋白沉淀和酸化后,以低浓度2-溴-对硝基苯乙酮为衍生化试剂,三乙胺为催化剂,在超声辐射下进行衍生化反应。色谱柱为Bennetnach C_(18)柱,流动相为甲醇-水(79∶21),柱温:25℃,流速:为1ml·min~(-1),检测波长:265nm。结果丙戊酸钠血药浓度在5~160μg·ml~(-1)范围内与峰面积呈良好线性关系(r=0.9995),平均回收率为94.8%,日内与日间RSD≤8.7%。结论本法可简便、快速、准确地测定癫痫患儿血清中丙戊酸钠浓度,适用于临床常规监测需要。
Objective To optimize the method for the determination of sodium valproate in the serum of children with epilepsy. Methods Cyclohexanecarboxylic acid was used as the internal standard. The blood samples were precipitated with acetonitrile and acidified, and then derivatized with low concentration of 2-bromo-p-nitroacetophenone as the derivatization reagent and triethylamine as catalyst under ultrasonic irradiation The reaction. The column was Bennetnach C_ (18) column. The mobile phase was methanol-water (79:21). The column temperature was 25 ℃, the flow rate was 1 ml · min -1 and the detection wavelength was 265 nm. Results The plasma concentration of valproate showed a good linear relationship with the peak area in the range of 5 ~ 160μg · ml ~ (-1) (r = 0.9995). The average recovery was 94.8%. The intraday and interday RSD were less than or equal to 8.7%. Conclusion This method can be simple, rapid and accurate determination of serum sodium valproate concentrations in children with epilepsy, suitable for routine clinical monitoring needs.
