研制不同孔径的硅胶,并进行了C_(18)烷基化学键合。探讨了硅胶孔径对蛋白质的保留、分离和回收率的影响。实验结果表明,硅胶孔径对蛋白原分离的柱效率有重要影响。然而,对本实验中绝大多数蛋白质,在不同孔径柱上,其保留时间基本保持不变。蛋白质的回收率随硅胶孔径增大而有所提高。此外,尚考察了流动相中有机溶剂的浓度对牛血清白蛋白(BSA)保留的影响。
The silica gel with different pore sizes was prepared and C 18 alkyl chemical bonding was carried out. The effects of silica gel pore size on protein retention, separation and recovery were discussed. The experimental results show that the pore size of silica gel has an important effect on the efficiency of column separation of protein. However, for most of the proteins in this experiment, the retention time remained basically unchanged on different pore size columns. The recovery of protein increased with the increase of silica gel pore size. In addition, the influence of the organic solvent concentration in the mobile phase on the retention of bovine serum albumin (BSA) was examined.
