目的:建立测定中药复方四物汤中主要成分含量的分析方法,比较四物汤共煎液与单煎混合液中没食子酸、芍药内酯苷、芍药苷、阿魏酸和苯甲酸的含量差异。方法:采用HPLC方法对四物汤共煎液与单煎混合液中的上述5个主要成分进行含量测定。色谱柱为Zorbax SB C_(18)(5μm,250mm×4.6mm)。流动相:0.01%(v/v)磷酸水溶液(A)-乙腈(B)梯度洗脱,洗脱程序为:0-7min,A-B保持95:5不变;7-12min,A-B由95:5线性变化至83:17;12-25min,A-B保持不变;25-27min,A-B由83:17线性变化至80:20;27-35min,A-B保持不变。流速:1.0mL·min~(-1);检测波长:230nm;柱温:25℃,进样量:10μL。结果:这5个化合物在测定的范围内表现出良好的线性(r>0.9995),方法的回收率在94.8%-103.1%之间。四物汤共煎液中没食子酸、芍药内酯苷、芍药苷、阿魏酸的含量均高于单煎混合液,而苯甲酸在四物汤共煎液中的含量低于单煎混合液。结论:四物汤共煎可提高有效成分的溶出,降低有毒成分的含量。
OBJECTIVE: To establish an analytical method for determining the content of major components in traditional Chinese medicine compound Siwu Decoction, and to compare the differences in contents of gallic acid, paeoniflorin, paeoniflorin, ferulic acid and benzoic acid in the decoction and single decoction of Siwu Tang. . Methods: The contents of the above five main components in the decoction and single decoction of Siwu Tang were determined by HPLC. The column was Zorbax SB C_(18) (5 μm, 250 mm×4.6 mm). Mobile phase: gradient elution of 0.01% (v/v) phosphoric acid aqueous solution (A)-acetonitrile (B), elution procedure: 0-7 min, AB retention 95:5 unchanged; 7-12 min, AB from 95:5 Linear change to 83:17; 12-25min, AB remained unchanged; 25-27min, AB linearly changed from 83:17 to 80:20; 27-35min, AB remained unchanged. Flow rate: 1.0 mL·min -1; Detection wavelength: 230 nm; Column temperature: 25°C, Sample injection volume: 10 μL. RESULTS: The five compounds showed good linearity (r>0.9995) within the range of determination. The recoveries of the method ranged from 94.8% to 103.1%. The contents of gallic acid, paeoniflorin, paeoniflorin and ferulic acid in the decoction of Siwu Tang were all higher than those in the single decoction, while the content of benzoic acid in the decoction of Siwu Tang was lower than that of the single decoction. . Conclusion: Simultaneous decoction of Siwu Tang can increase the dissolution of active ingredients and reduce the content of toxic components.
