采用荧光光谱法研究了β-环糊精(β-CD)与依托泊苷(VP-16)之间的超分子包结作用。考察了时间、温度和浓度对包结反应的影响。并用紫外-可见光光谱法对包结物溶液进行了表征。实验结果表明:VP-16本身具有天然荧光,与β-CD形成1∶1的超分子包结物后,荧光强度增大,β-CD对VP-16有荧光增敏作用。在22℃,pH7.0时,包结常数为2.63×105L·mol-1。根据VP-16在316nm处发射强荧光,建立了定量检测微量VP-16的荧光分析法,其线性良好,相关系数为0.9999。
Fluorescence spectroscopy was used to study the supramolecular inclusion of β-cyclodextrin (β-CD) and etoposide (VP-16). The effects of time, temperature and concentration on the entrapment reaction were investigated. The inclusion complex solution was characterized by UV-Vis spectroscopy. The results showed that VP-16 itself has a natural fluorescence, and after forming a 1: 1 supramolecular inclusion complex with β-CD, the fluorescence intensity increases, and β-CD has a fluorescence sensitizing effect on VP-16. At 22 ℃, pH7.0, the inclusion constant was 2.63 × 105L · mol-1. Based on the strong fluorescence of VP-16 emission at 316 nm, a fluorescence assay for quantitative determination of VP-16 was established. The linearity was good with a correlation coefficient of 0.9999.
