目的:研究植物雌激素刺芒柄花素对去卵巢+高脂血症大鼠主动脉弓雌激素受体β(ERβ)表达的影响以了解刺芒柄花素对去势大鼠心血管的保护作用。方法:实验选取SD大鼠制作去势高脂模型,以Western blot和real-timePCR方法检测主动脉弓中ERβ的含量。结果:Western blot结果表明模型组大鼠主动脉弓ERβ的含量较假手术组明显减少,尼尔雌醇组和高剂量刺芒柄花素组较模型组明显升高(P<0.05),低剂量刺芒柄花素组较模型组也升高,但两者差异无统计学意义。real-time结果显示尼尔雌醇组ERβ含量最高,高剂量刺芒柄花素组次之,低剂量略低,模型组大鼠ERβ含量最低。结论:雌激素水平降低可使大鼠主动脉血管雌激素受体β表达减少,合适剂量的刺芒柄花素能上调去势大鼠主动脉血管ERβ的表达
Objective: To study the effect of phytoestrogen prothromin on the expression of estrogen receptor β (ERβ) in the aortic arch of ovariectomized rats and hyperlipidemic rats to understand the protective effect of prothymidine on cardiovascular in ovariectomized rats . METHODS: SD rats were selected to make castrated hyperlipidemic model. The contents of ERβ in the aortic arch were detected by Western blot and real-time PCR. Results: Western blot results showed that the content of ERβ in the aortic arch of rats in the model group was significantly decreased compared with that in the sham-operation group. The levels of nylestriol and high-intensity stinger-formononetin in model group were significantly higher than those in model group (P <0.05) Formononetin group than the model group also increased, but the difference was not statistically significant. The results of real-time showed that the highest level of ERβ was found in nilestriol group, followed by the high-dose thymus fonaton group, and the low dose was slightly lower. The content of ERβ in model group was the lowest. CONCLUSION: The decrease of estrogen can decrease the expression of estrogen receptor β in the aorta of rats, and the appropriate dose of thiamethoxol can up-regulate the expression of ERβ in the aorta of ovariectomized rats
