目的 探讨人工合成的 Cp G ODN作为佐剂对乙肝病毒基因疫苗诱导小鼠产生抗 HBs的影响 .方法 构建编码乙型肝炎病毒 (HBV)表面抗原蛋白 S的重组真核表达质粒p CR3.1- S作为 HBV基因疫苗 ,人工合成含 Cp G motif的硫代磷酸寡核苷酸作为佐剂 ,以 BAL B/ c小鼠作为实验动物进行免疫接种 ,采用 EL ISA法检测免疫小鼠的抗 HBs应答 .结果 与生理盐水作为佐剂组相比较 ,应用 Cp G ODN组小鼠产生更强的抗 HBs反应 (P<0 .0 5 ) .结论 Cp G ODN作为佐剂可促进 HBV基因疫苗诱导 BAL B/ c小鼠产生抗 HBs应答 .
Objective To investigate the effect of synthetic CpG ODN as an adjuvant on the anti-HBs production in mice induced by hepatitis B virus gene vaccine.Methods To construct a recombinant eukaryotic expression plasmid pCR3.1- S as an HBV gene vaccine, phosphorothioate oligonucleotides containing CpG motif were synthesized as an adjuvant, BALB / c mice were immunized as experimental animals, and the anti-HBs response of immunized mice was detected by EL ISA Results Compared with saline as an adjuvant, mice immunized with CpG ODN produced a stronger anti-HBs response (P <0.05) .Conclusion CpG ODN as adjuvant can promote the induction of BALB / c mice develop anti-HBs responses.
