目的:采用限制片段差异显示聚合酶链反应(RFDD-PCR)建立卵巢癌组织与正常卵巢组织基因差异表达谱,分析卵巢癌相关基因的表达差异。方法:采集上皮性卵巢癌手术患者的卵巢癌组织和正常卵巢组织,用RFDD-PCR建立卵巢癌组织与正常卵巢组织基因差异表达谱,对各组织间有表达差异的基因进行生物信息学分析,筛选与卵巢癌发病机制相关的基因。结果:构建了两种不同组织的基因表达谱,获得有意义的电泳条带5642条,其中卵巢癌组织2227条,正常卵巢组织3384条,癌组织和正常组织间有表达差异的片段为2082个,占表达数的36.9%。结论:利用RFDD-PCR差异显示技术能够建立卵巢癌组织和正常卵巢组织基因差异表达谱。
OBJECTIVE: To establish differentially expressed genes profiles of ovarian cancer tissues and normal ovarian tissues by restriction fragment PCR (RFDD-PCR) and to analyze the expression differences of ovarian cancer-related genes. Methods: The ovarian cancer tissues and normal ovarian tissues of patients with epithelial ovarian cancer were collected. Gene differential expression profiles of ovarian cancer tissues and normal ovarian tissues were established by RFDD-PCR. Bioinformatic analysis was performed on the genes with different expression among the tissues. Screening of genes associated with the pathogenesis of ovarian cancer. Results: Gene expression profiles of two different tissues were constructed and 5642 bands were found to be meaningful. Among them, 2227 ovarian cancer tissues, 3384 normal ovarian tissues and 2082 non-cancerous and normal tissues , Accounting for 36.9% of the total number of expressions. Conclusion: Gene differential expression profiling of ovarian cancer tissue and normal ovary tissue can be established by RFDD-PCR differential display technique.
