RCRF在高级别肾癌组织中表达上调并影响肾癌患者预后

目的研究不同病理级别肾细胞癌中差异表达的长链非编码RNAs(lncRNAs),找到其中一种lncRNA——RCRF与肾癌患者临床病理资料的关系,从而判断肾癌预后。方法取6例肾细胞癌原发灶肿瘤组织,3例病理分级为FuhrmanⅠ级,另3例为FuhrmanⅢ~Ⅳ级,采用lncRNA芯片技术,筛选差异表达的lncRNAs。另取40例肾细胞癌患者的癌组织及癌旁正常组织,22例病理分级为FuhrmanⅠ~Ⅱ(低级别),18例为FuhrmanⅢ~Ⅳ(高级别),利用实时定量PCR技术检测在癌及癌旁非肿瘤组织中RCRF的表达,并研究RCRF与临床分期、病理分级、淋巴结转移、远处转移之间的关系。结果筛选出320个与肾细胞癌病理级别相关的差异表达lncRNAs。肾癌组织尤其是高级别肾癌组织中RCRF的相对表达上调(P<0.01)。RCRF表达与肾癌病理分级、淋巴转移及远处转移有关(P<0.05)。结论 RCRF在高级别肾癌组织中表达上调,并且影响肾癌患者的预后。 Objective To study the relationship between lncRNA - RCRF and the clinicopathological data of patients with renal cell carcinoma (RCC), and to determine the prognosis of renal cell carcinoma (RCC) by differentially expressed long - chain non - coding RNAs (lncRNAs) in different pathological grades of renal cell carcinoma. Methods Six primary renal cell carcinoma tissues were obtained. The pathological grade was Fuhrman grade Ⅰ and the third grade was Fuhrman grade Ⅲ ~ Ⅳ. The lncRNA microarray was used to screen differentially expressed lncRNAs. Another 40 patients with renal cell carcinoma and adjacent normal tissue, 22 cases of pathological grade Fuhrman Ⅰ ~ Ⅱ (low level), 18 cases of Fuhrman Ⅲ ~ Ⅳ (high level), using real-time quantitative PCR detection of cancer and The expression of RCRF in noncancerous adjacent tissues and the relationship between RCRF and clinical stage, pathological grade, lymph node metastasis and distant metastasis were studied. Results A total of 320 differentially expressed lncRNAs were screened for pathological grade of renal cell carcinoma. The relative expression of RCRF was significantly up-regulated in renal cancer tissues, especially in high-grade renal carcinoma (P <0.01). The expression of RCRF was correlated with pathological grade, lymph node metastasis and distant metastasis (P <0.05). Conclusion RCRF is upregulated in high grade renal cell carcinoma and affects the prognosis of patients with renal cell carcinoma.
