目的探讨类风湿关节炎(RA)关节液和滑膜组织中人诱骗受体3(DcR3)表达水平变化,以及关节滑膜组织炎性细胞数与DcR3表达水平的相关性。方法采用免疫细胞化学法,扫描分析了30例RA患者、20例骨关节炎(OA)患者和30例关节正常的骨折患者滑膜组织中DcR3阳性细胞,并计数其中的炎性细胞;用酶联免疫吸附试验测定30例RA患者和20例OA患者关节液中DcR3浓度,将试验数据进行统计学处理。结果 RA患者DcR3在滑膜细胞中高表达,阳性率约72%,并且与炎性细胞数呈正相关(r=0.989,P<0.01);RA患者关节液DcR3浓度显著高于OA患者,差异具有统计学意义(P<0.01)。结论 RA患者滑膜组织和关节液中DcR3高表达可能与自身免疫性炎症反应有关,DcR3可能参与类风湿关节炎的发病机制。
Objective To investigate the expression of human decoy receptor 3 (DcR3) in synovial fluid and synovial tissue of patients with rheumatoid arthritis (RA) and the correlation between the expression of DcR3 and inflammatory cells in synovial tissue. Methods Immunocytochemistry was used to scan DcR3 positive cells in synovial tissue of 30 patients with RA, 20 patients with osteoarthritis (OA) and 30 patients with normal joint fracture. The inflammatory cells were counted. The concentration of DcR3 in the synovial fluid of 30 patients with RA and 20 patients with OA was measured by ELISA, and the experimental data were statistically analyzed. Results DcR3 was highly expressed in synovial cells in RA patients, with a positive rate of 72% and a positive correlation with inflammatory cells (r = 0.989, P <0.01). DcR3 concentrations in synovial fluid of RA patients were significantly higher than those in OA patients Significance (P <0.01). Conclusion The high expression of DcR3 in synovial tissue and synovial fluid of RA patients may be related to the autoimmune inflammatory reaction. DcR3 may be involved in the pathogenesis of rheumatoid arthritis.
