目的比较卫矛提取物不同极性组分总黄酮含量及抗氧化抗炎活性。方法 60%乙醇提取卫矛黄酮类化合物,经萃取制得4个不同极性的组分(石油醚组分、乙酸乙酯组分、正丁醇组分、水组分)。采用分光光度法测定各组分的总黄酮含量;通过清除DPPH自由基、ABTS自由基试验和还原力试验,评价各组分的抗氧化活性;利用脂多糖诱导小鼠腹腔巨噬细胞RAW264.7释放一氧化氮模型,评价各组分的抗炎活性。结果卫矛提取物总黄酮的含量为(271.25±5.51)mg·g-1,其中乙酸乙酯组分的总黄酮含量最高,为(773.61±12.03)mg·g-1,显著高于总提取物和其他组分(P<0.01)。活性评价结果表明卫矛提取物具有良好的抗氧化和抗炎活性,其中乙酸乙酯组分的抗氧化活性最强,乙酸乙酯组分和石油醚组分则具有良好的抗炎活性。结论卫矛的化学成分具有良好的多样性,其作用机制具有多组分、多靶点、多途径的特点,其具体机制有待进一步深入研究。
Objective To compare the content of total flavonoids in different polar fractions and the anti-oxidation and anti-inflammatory activity of the extract. Methods Flavonoids of Pueraria baicalensis were extracted by 60% ethanol, and four fractions of different polarity (petroleum ether, ethyl acetate, n-butanol and water) were extracted. The content of total flavonoids in each component was determined by spectrophotometry. The antioxidant activity of each component was evaluated by scavenging DPPH free radical, ABTS free radical test and reducing power test. Lipopolysaccharide was used to induce RAW 264.7 Nitric oxide model was released to evaluate the anti-inflammatory activity of each component. Results The content of total flavonoids in Euonymus fortunei extract was (271.25 ± 5.51) mg · g-1, and the highest content of total flavonoids in ethyl acetate fraction was (773.61 ± 12.03) mg · g-1, which was significantly higher than that of total extraction And other components (P <0.01). The results of the activity evaluation showed that the Euonymus fortunei extract has good anti-oxidation and anti-inflammatory activity, of which the ethyl acetate component has the strongest antioxidant activity, and the ethyl acetate component and the petroleum ether component have good anti-inflammatory activity. Conclusion The chemical composition of Euonymus fortunei has a good diversity, its mechanism of action with multi-component, multi-target, multi-path characteristics, the specific mechanism needs further study.
