本试验主要从液体培养基的种类、培养温度、培养方式、培养天数、接种量和培养基的pH值及碳源等几个方面入手研究了大豆灰斑病菌最佳的产毒条件。利用大豆胚根伸长抑制率和单复叶萎蔫指数来测定毒素的产量。实验结果得出该菌的最佳产毒条件是利用Czapek 培养基,以蔗糖作为碳源,糖含量为3% ,将培养基的pH调到6- 7,接种三块直径为6m m 的新鲜菌丝块,于25- 28℃下静止培养25- 27 天。
This experiment mainly studied the optimum conditions of toxin production of Cercospora sojina from the aspects of liquid medium type, culture temperature, culture method, culture days, inoculum size, pH value of culture medium and carbon source. Toxoplasma production was determined using the inhibition rate of soybean root elongation and the single leaf wilting index. The experimental results showed that the optimal conditions of producing bacteria were as follows: Czapek medium was used, sucrose was used as carbon source, the content of sugar was 3%, the pH of the medium was adjusted to 6-7, and three fresh Mycelium, at 25-28 ℃ static culture 25-27 days.
