目的:观察中药何首乌饮对衰老模型大鼠卵巢形态学的影响。方法:将48只SD大鼠随机分为3组:正常组、模型组、预防组。采用D-半乳糖连续腹腔注射法制造雌性大鼠亚急性衰老模型,预防组在造模的同时以中药方剂何首乌饮灌胃。应用HE染色及透射电镜方法观察各组大鼠卵巢组织形态学的变化。结果:何首乌饮明显增加了衰老大鼠卵巢组织中原始卵泡、生长卵泡及黄体的数量(P<0.05,P<0.01),且明显改善了衰老大鼠卵巢颗粒细胞的超微结构,使滑面内质网增加,线粒体及高尔基复合体丰富。结论:何首乌饮通过影响衰老大鼠卵巢形态学的改变,具有预防和延缓颗粒细胞凋亡而达到阻止卵泡闭锁,可能是抗大鼠性腺衰老的作用机制之一。
Objective: To observe the effect of Heshouwuyin, a traditional Chinese medicine, on the morphology of ovaries of aging rats. Methods: Forty-eight SD rats were randomly divided into three groups: normal group, model group, and prevention group. The subacute aging model of female rats was made by continuous intraperitoneal injection of D-galactose. The prevention group was given intragastric administration of the traditional Chinese medicine prescription Polygonum multiflorum at the same time as the model building. HE staining and transmission electron microscopy were used to observe the morphological changes of ovarian tissues in each group. RESULTS: Heshouwuyin significantly increased the number of primordial follicles, growing follicles and corpus luteum in the ovary of aging rats (P<0.05, P<0.01), and significantly improved the ultrastructure of granulosa cells in aging rats, resulting in a smooth surface. The endoplasmic reticulum increased, and mitochondria and Golgi complexes were abundant. Conclusion: Heshouwuyin can prevent and delay follicular atresia by preventing and delaying the apoptosis of granulosa cells through affecting the morphological changes of the ovary of aging rats, which may be one of the mechanisms of anti-senility of rat gonads.
